Business
Applied Genetic Technologies Corporation Applied Genetic Technologies Corporation (AGTC) is a clinical-stage biotechnology company focused on developing proprietary gene therapies for patients with rare and debilitating ophthalmologic, otologic, and central nervous system diseases. The company offers a pipeline of product candidates targeting inherited retinal diseases, including AGTC-501 (laruparetigene zosaparvovec) for X-linked retinitis pigmentosa (XLRP), programs for achromatopsia (ACHM CNGB3 and ACHM CNGA3 mutations), X-linked retinoschisis (XLRS), blue cone monochromacy, wet age-related macular degeneration, and optogenetics-based therapies for advanced retinal disease; it also advances preclinical candidates for alpha-1 antitrypsin deficiency, adrenoleukodystrophy, otology indications such as genetic deafness, and central nervous system disorders including frontotemporal dementia and amyotrophic lateral sclerosis. AGTC leverages expertise in adeno-associated virus (AAV) vector design, synthetic promoters, expression cassettes, and manufacturing to create customized therapies with high unmet medical need, clinical feasibility, and commercial potential, primarily serving patients with orphan genetic diseases through subretinal and intravitreal delivery methods. Founded in 1999 and headquartered in Alachua, Florida, with additional operations in Cambridge, Massachusetts, the company operates in the United States and collaborates with partners like Bionic Sight for optogenetics and Otonomy for neurotology. In a major strategic shift, AGTC was acquired in December 2022 by a Syncona Limited portfolio company through a tender offer valued at approximately $23.5 million upfront plus up to $50 million in contingent value rights, after which its lead asset AGTC-501 and broader pipeline were integrated into Beacon Therapeutics, a new ophthalmic gene therapy entity launched in June 2023 with £96 million ($120 million) in funding from Syncona and others including Oxford Science Enterprises. Beacon Therapeutics, building on AGTC's programs, raised an additional $170 million in Series B financing in 2024 led by Forbion to advance development, and reported positive interim data from Phase 2 DAWN and SKYLINE trials of laru-zova (formerly AGTC-501) in 2025, showing sustained visual function improvements including low luminance visual acuity and microperimetry in XLRP patients; enrollment completed in the pivotal Phase 2/3 VISTA trial with topline data expected in the second half of 2026. These changes reflect AGTC's transition from independent public operations (delisted from Nasdaq post-acquisition) to a supported platform within Beacon's focus on rare and prevalent blinding ocular diseases like XLRP, cone-rod dystrophy, and dry AMD.
