Phio Pharmaceuticals Corp. is a clinical-stage biotechnology firm focused on advancing immuno-oncology therapies within the United States. The company’s foundation rests upon its proprietary INTASYL™ platform, which engineers self-delivering RNAi compounds designed to silence genes that enable tumors to evade the body’s immune system. This innovative approach seeks to enhance the immune response in cancerous environments and is adaptable for both intratumoral and adoptive cell therapy applications. Among its pipeline candidates, PH-762 is an INTASYL compound engineered to diminish the expression of the PD-1 checkpoint protein, thereby activating immune cells to identify and eliminate cancer cells. This compound is currently undergoing Phase 1b clinical trials for various cutaneous malignancies, including advanced squamous cell carcinoma, melanoma, and Merkel cell carcinoma, and has recently shown encouraging safety and efficacy results in these studies. PH-762 is also being explored in conjunction with AgonOx Inc.'s "double positive" tumor-infiltrating lymphocyte technology to improve T cell-based cancer immunotherapies. Another key program, PH-894, targets the epigenetic protein BRD4 to influence gene expression in both immune and tumor cells, aiming to bolster the effectiveness of adoptive cell therapies. Additionally, PH-804 is under development to target the TIGIT immune receptor, enhancing the function of T cells and natural killer cells. The company is also investigating other INTASYL compounds, including one targeting CTLA-4, and has expanded its focus to include potential cosmeceutical applications through an exploratory collaboration. Phio Pharmaceuticals Corp. was originally established in 2011 as RXi Pharmaceuticals Corporation, later adopting its current name in November 2018 to reflect its dedicated pursuit of immuno-oncology treatments.
