Blueprint Medicines Corporation, now operating as a Sanofi company, is a biopharmaceutical firm focused on developing precision therapies for genomically defined cancers and rare immunological conditions across the globe. The company offers AYVAKIT®/AYVAKYT® (avapritinib), a kinase inhibitor approved for various indications including adults with unresectable or metastatic gastrointestinal stromal tumors (GIST) harboring a PDGFRA exon 18 mutation, advanced systemic mastocytosis (AdvSM), and indolent systemic mastocytosis (ISM) with moderate to severe symptoms. Its portfolio also historically included GAVRETO® (pralsetinib), an oral precision therapy targeting RET fusion-positive non-small cell lung cancer, RET-mutant medullary thyroid cancer, and RET fusion-positive thyroid cancer; however, the U.S. rights for GAVRETO were transitioned to Rigel Pharmaceuticals in February 2024. Within its robust pipeline, Blueprint Medicines is progressing elenestinib (BLU-263), a next-generation KIT D816V inhibitor currently in a registration-enabling Phase 3 trial for indolent systemic mastocytosis. Another significant candidate is BLU-808, a highly selective oral wild-type KIT inhibitor undergoing Phase 2 evaluation for chronic inducible urticaria and chronic spontaneous urticaria, with broader potential across immunological diseases. Additionally, fidrisertib (formerly BLU-782), an ALK2 inhibitor, is licensed exclusively worldwide to Clementia Pharmaceuticals, a subsidiary of Ipsen, for fibrodysplasia ossificans progressiva. The company further expanded its scientific platform to include targeted protein degradation through a collaboration with VantAI, which was recently amended in May 2025 to advance novel therapies. Blueprint Medicines was initially incorporated in October 2008 and adopted the name Blueprint Medicines Corporation in June 2011. The enterprise maintains its headquarters in Cambridge, Massachusetts. In a significant strategic move, Sanofi entered into an agreement to acquire Blueprint Medicines Corporation for up to $9.5 billion in June 2025, thereby integrating its precision medicine capabilities into Sanofi's rare immunology and oncology portfolios.
