Septerna, Inc. (Nasdaq: SEPN) is a clinical-stage biotechnology company that discovers and develops oral small molecule therapeutics targeting G protein-coupled receptors (GPCRs) using its proprietary, industrial-scale Native Complex Platform. The company advances a wholly owned pipeline focused initially on endocrinology, immunology and inflammation, and metabolic diseases, including SEP-479, an oral small molecule agonist of the parathyroid hormone 1 receptor (PTH1R) for hypoparathyroidism; SEP-631, an oral small molecule negative allosteric modulator (NAM) of MAS-related G protein-coupled receptor X2 (MRGPRX2) for chronic spontaneous urticaria and other mast cell-driven diseases; and a TSHR program featuring an oral small molecule NAM of the thyroid-stimulating hormone receptor (TSHR) for Graves' disease and thyroid eye disease. Additional research areas span neurology, women's health, cardiovascular disease, and respiratory disease; partnered metabolic programs target obesity, type 2 diabetes, and other cardiometabolic conditions via small molecule agonists of GLP-1R, GIPR, and GCGR receptors. Septerna serves patients with significant unmet needs across these therapeutic segments through novel mechanistic approaches, including agonists, antagonists, and allosteric modulators enabled by biomarker-driven strategies. Founded in 2019 and headquartered in South San Francisco, California, the company--formerly known as GPCR NewCo, Inc., with a name change in June 2021--went public on Nasdaq in October 2024. In a major strategic development, Septerna entered a global collaboration with Novo Nordisk in May 2025 to discover, develop, and commercialize multiple oral small molecule medicines for obesity, type 2 diabetes, and other cardiometabolic diseases targeting specified GPCR targets including GLP-1, GIP, and glucagon receptors; the agreement provides over $200 million in upfront and near-term payments, potential milestones exceeding $2.2 billion, tiered royalties on global net sales, and an option for Septerna to participate in worldwide profit-sharing for one program, with Novo Nordisk funding all research and development expenses post-IND-enabling activities for partnered programs. Recent pipeline progress includes dosing of the first participants in the Phase 1 clinical trial of SEP-631 in 2025 and advancement of SEP-479 and TSHR programs toward clinical stages, alongside reporting third quarter 2025 financial results that highlight a cash position extended beyond early 2028. Septerna operates primarily from the United States with no disclosed subsidiaries or parent entities.
