(Nasdaq: AGEN), a clinical-stage immuno-oncology company founded in 1994 and headquartered in Lexington, Massachusetts, discovers, manufactures, and develops therapies that activate the immune system against cancer and infectious diseases. The company focuses on immune checkpoint modulating antibodies including botensilimab, a next-generation CTLA-4 antibody with enhanced Fc functionality, balstilimab, a PD-1 inhibitor, zalifrelimab, a CTLA-4 antibody, and earlier-stage candidates such as AGEN1571 (anti-ILT2), AGEN1777 (TIGIT bispecific), AGEN2373 (CD137), and AGEN1423 (CD73-TGFβ); adoptive cell therapies through its subsidiary MiNK Therapeutics, Inc. (Nasdaq: INKT); and vaccine adjuvants via SaponiQx, Inc., featuring proprietary QS-21 from cultured plant cells and platforms like Retrocyte Display for antibody discovery and AdenoJuice for personalized vaccines. Agenus operates primarily in the United States and Europe, with a pipeline targeting solid tumors such as metastatic colorectal cancer, cervical cancer, and other treatment-resistant malignancies, supported by in-house cGMP manufacturing capabilities. In recent developments, Agenus entered a $141 million strategic collaboration with Zydus Lifesciences in June 2025, exchanging its biologics CMC facilities in Emeryville and Berkeley, California, for $75 million upfront plus up to $50 million in contingent payments, alongside a $16 million equity investment from Zydus and an exclusive manufacturing agreement for botensilimab/balstilimab (BOT/BAL) to support clinical and commercial scale-up. Earlier, in May 2024, the company secured a $100 million royalty financing agreement with Ligand Pharmaceuticals, with potential syndication up to $200 million, to fund BOT/BAL's Phase 3 trial in relapsed/refractory microsatellite stable colorectal cancer and launch activities. Additional advancements include completing enrollment in a Phase 2 BOT/BAL trial in advanced MSS colorectal cancer, receiving Fast Track Designation, launching a Q4 2025 international trial across Canada, France, Australia, and New Zealand, and appointing Dr. José Iglesias as Chief Medical Affairs Officer to expand global access programs.
