Business
Foghorn Therapeutics Inc., a biotechnology firm in clinical development, focuses on discovering and advancing therapeutic agents that target genetically defined vulnerabilities within the intricate chromatin regulatory system. The company leverages its proprietary Gene Traffic Control® platform to identify, validate, and develop potential drug candidates within this essential cellular control mechanism. Currently, Foghorn is progressing several programs. Its lead candidate, FHD-909, a first-in-class oral selective SMARCA2 inhibitor, is currently in a Phase 1 trial for SMARCA4 mutated cancers, primarily targeting non-small cell lung cancer. Additionally, the company is advancing other programs, including selective CBP degrader and EP300 degrader programs toward Investigational New Drug (IND) applications, with preclinical data supporting their potential, and a selective ARID1B degrader program. Previously, Foghorn investigated FHD-286, an inhibitor of BRG1 and BRM. While some activity was observed in metastatic uveal melanoma, the company has decided not to advance it independently for this indication. For relapsed or refractory acute myeloid leukemia and myelodysplastic syndrome, a combination study for FHD-286 was initiated, with recent data from a Phase 1 study indicating antileukemic activity despite no objective responses, suggesting further combination investigation. Another program, FHD-609, a BRD9 protein degrader for synovial sarcoma and SMARCB1-deleted tumors, saw its clinical study enrollment paused and was placed on partial clinical hold by the FDA due to a safety event. Foghorn maintains a strategic partnership with Loxo Oncology at Eli Lilly and Company to co-develop novel oncology therapies, including a selective BRM program and other discovery-stage targets. The company was founded in 2015 and operates from Cambridge, Massachusetts.
